چکیده

The hormonal derivatives diethylstilbestrol, hexestrol, dienestrol, cortisone, estradiol-17a, estradiol-17/3, progesterone, and phloretin were compared with regard to their capacity to enhance carcinostasis in the presence of the niacin antagonist 6aminonicotinamide. Diethylstilbestrol and hexestrol were found to be more powerful than either estradiol-17/îor dienestrol. Phloretin, estradiol-17a, and progesterone were without activity. Cortisone, either alone or in the presence of 6-aminonicotinamide, exhibited slight antitumor activity. In the absence of 6-aminonicotinamide, diethylstilbestrol, hexestrol, dienestrol, and estradiol-17/3 had no antitumor activity. The suggestive correlation between estrogenic activity and enhancement of carcino stasis in the presence of 6-aminonicotinamide is discussed.

Quantitative biochemical differences between tumor and host tissue. VI. 6-Aminonicotinamide antagonism of DPN-dependent enzymatic systems.

The general mechanism whereby a tumor, an integral part of the host, may be selectively de stroyed without permanently altering the other tissues of the host is unknown. Extensive research has been directed toward finding something unique in tumor metabolism. These studies indicate that the same metabolic pathways and metabolites found in tumor tissue are also found in other host tissues; i.e.,...

Biochemical modulation of tumor cell energy: regression of advanced spontaneous murine breast tumors with a 5-fluorouracil-containing drug combination.

This report describes a highly active chemotherapeutic drug combination, consisting of N-(phosphonacetyl)-L-aspartate plus 6-methylmercaptopurine riboside plus 6-aminonicotinamide plus 5-fluorouracil, in CD8F1 mice bearing spontaneous, autochthonous, breast tumors or first-passage advanced transplants of these spontaneous tumors. The combination and sequence of administration of these drugs wer...

Enhancement of random migration and chemotactic response of human leukocytes by ascorbic acid.

Incubation of human leukocytes with ascorbic acid at neutral pH and at concentrations 10-50 times that of normal blood levels augmented both the in vitro random migration and chemotaxis of the cells by 100-300% without influencing their phagocytic capacity. Enhancement of mobility by ascorbate was evident for isolated neutrophils, eosinophils, and mono-nuclear leukocytes and was independent of ...

Clinical experience with 6-aminonicotinamide.

The toxicity of 6-Aminonicotinamide in 60 patients with advanced malignant disease has been described. The toxicity took two clinical forms: a "deficiency" state with char acteristics of a mixed B-complex vitamin deficiency, and a neurologic disturbance pre sumably due to a direct action on the central nervous system. Individual susceptibility was widely variable. 6-Aminonicotinamide exhibited ...

Synergistic antileukemic effect of 6-aminonicotinamide and 1,3-bis(2-chloroethyl)-1-nitrosourea on L1210 cells in vitro and in vivo.

A series of nicotinamide analogs were evaluated for their ability to inhibit L1210 cell poly(adenosine diphosphoribose) polymerase, and also for their ability to potentiate the cytocidal effects of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU), nitrogen mustard, and gamma-irradiation. In L1210 cells growing in culture and in vivo. In vitro, nicotinamide, 5-methylnicotinamide, 6-aminonicotinamide,...
